Mouse IP-10 ELISA Kit (CXCL10) (ab214563) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of IP-10 (CXCL10) protein in cell culture supernatant, edta plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse IP-10 (CXCL10) with 3.4 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. ASSAY SPECIFICITY
This kit recognizes both native and recombinant mouse IP-10 protein in serum, plasma (EDTA), and cell culture supernatant.
Cell and tissue extract samples have not been tested with this kit.
CROSS REACTIVITY
Recombinant human IP-10 was prepared at 4,000 pg/mL and 400 pg/mL and assayed for cross reactivity. No cross-reactivity was observed.
SPECIES REACTIVITY
This kit recognizes mouse IP-10 protein.
Other species reactivity was determined by measuring 50% serum samples of various species, interpolating the protein concentrations from the mouse standard curve, and expressing the interpolated concentrations as a percentage of the protein concentration in mouse serum assayed at the same dilution.

C-X-C motif chemokine 10 (CXCL10 or IP-10) is a small 10.8kD protein that is secreted by several cell types in response to interferon-gamma (IFNg). These cell types include monocytes, endothelial cells and fibroblasts. Upon secretion, CXCL10 is cleaved into an 8.7kD biologically active protein to function in chemotaxis for T-cells, NK cells, monocytes/macrophages and dendritic cells. In addition, CXCL10 has antitumor activity through the inhibition of bone marrow colony formation and angiogenesis. CXCL10 elicits its effects by binding to the cell surface chemokine receptor 3 (CXCR3).

Interpolated concentrations of mouse IP-10 in RAW264.7 cells treated for 48 hours in the presence and absence of 5 µg/mL Lipopolysaccharide (LPS).The concentrations of IP-10 were measured in duplicates, interpolated from the IP-10 standard curves and corrected for sample dilution. Undiluted samples are as follows: 4% supernatant. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IP-10 concentration was determined to be 21.7 ng/mL in in RAW 264.7 supernatant.

Native IP-10 was measured in duplicate in 50% serum and 50% EDTA plasma diluted in Sample Diluent 25BP and concentrations interpolated from a standard curve diluted in Sample Diluent 25BP. The interpolated dilution factor corrected values are graphed (mean +/- SD).

The MDD was determined by calculating the mean of zero standard replicates and adding 2 standard deviations then extrapolating the corresponding concentration.
