Mouse IL4R ELISA Kit (ab256403) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of IL4R protein in cell culture supernatant, cit plasma, edta plasma, hep plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse IL4R with 7.08 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
IL4R is a receptor for both interleukin 4 and interleukin 13. IL-4R is involved in the JAK1/2/3-STAT6 pathway. The IL4 response is essential in promoting Th2 differentiation and regulating IgE production, chemokine and mucus production at sites of allergic inflammation. In certain cell types, IL-4R can signal through activation of insulin receptor substrates, IRS1/IRS2. Soluble IL4R (sIL4R) inhibits IL4-mediated cell proliferation and IL5 upregulation by T-cells. Mouse IL-4R shares 52.2%, 76.0%, 50.7%, and 52.4% sequence homology with human, rat, bovine, and monkey respectively.

Function

Contains 1 copy of a cytoplasmic motif that is referred to as the immunoreceptor tyrosine-based inhibitor motif (ITIM). This motif is involved in modulation of cellular responses. The phosphorylated ITIM motif can bind the SH2 domain of several SH2-containing phosphatases. -
Post-translational
modificationsOn IL4 binding, phosphorylated on C-terminal tyrosine residues. Phosphorylation on any one of tyrosine residues, Tyr-575, Tyr-603 or Tyr-631, is required for STAT6-induced gene induction.
The soluble form (sIL4R/IL4BP) can also be produced by proteolytic cleavage at the cell surface (shedding) by a metalloproteinase. -

The IL4R standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of IL4R were measured in duplicates, interpolated from the IL4R standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 10%, plasma (citrate) 10%, plasma (EDTA) 10%, and plasma (heparin) 5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IL4R concentration was determined to be 14.42 ng/mL in serum, 9.87 ng/mL in plasma (citrate), 10.28 ng/mL in plasma (EDTA), 12.14 ng/mL in plasma (heparin).

Undiluted samples are as follows: spleen tissue culture supernatant 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IL4R concentration was determined to be 1465.50 pg/mL in spleen tissue culture supernatant.
