Mouse IL-3 ELISA Kit (ab222509) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of IL-3 protein in cit plasma, edta plasma, hep plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse IL-3 with 12 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
Interleukin 3 (IL-3) is a secreted 166 amino acids long, glycosylated interleukin coded by the IL3 gene. Previous names reported for IL-3 are Hematopoietic growth factor, Mast cell growth factor, multi-potential colony-stimulating factor, and P-cell-stimulating factor. IL-3 has been found to stimulate a variety of cell activities such as; proliferation, differentiation, survival of pluripotent hematopoietic stem cells, and may be associated with neurologic disorders. IL-3 is expressed by activated T-cells, mast cells, and natural killer cells. IL-3 performs its biological action by binding to a transmembrane receptor complex consisting of IL-3RA, also known as CD123, and a signal inducing beta subunit, which also acts as a receptor for colony stimulating factor 2 and Interleukin 5.

The IL-3 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of IL-3 were measured in duplicates, interpolated from the IL-3 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 50%, plasma (citrate) 50%, plasma (heparin) 50% and plasma (EDTA) 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).

Interpolated concentrations of native IL-3 in an unstimulated and PHAM (10 µg/mL PHA and 10 ng/mL PMA, 48 hr) stimulated EL4 cell culture supernatant samples
The concentrations of IL-3 were measured in duplicate and interpolated from the IL-3 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IL-3 concentration was determined to be 1834 pg/mL in neat stimulated EL4 culture supernatant samples and not detectable in unstimulated samples.
