Human XIAP ELISA Kit (ab234576) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of XIAP protein in cell culture extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human XIAP with 114.3 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Apoptotic suppressor. Has E3 ubiquitin-protein ligase activity. Mediates the proteasomal degradation of target proteins, such as caspase-3, SMAC or AIFM1. Inhibitor of caspase-3, -7 and -9. Mediates activation of MAP3K7/TAK1, leading to the activation of NF-kappa-B. -

Involvement in disease
Defects in XIAP are the cause of lymphoproliferative syndrome X-linked type 2 (XLP2) [MIM:300635]. XLP is a rare immunodeficiency characterized by extreme susceptibility to infection with Epstein-Barr virus (EBV). Symptoms include severe or fatal mononucleosis, acquired hypogammaglobulinemia, pancytopenia and malignant lymphoma. -

Domain
The first BIR domain is involved in interaction with TAB1/MAP3K7IP1 and is important for dimerization. The interactions with SMAC and PRSS25 are mediated by the second and third BIR domains. -

The concentrations of XIAP were measured in duplicate and interpolated from the XIAP standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean XIAP concentration was determined to be 8.2 ng/mL in sample and 18.2 ng/mL in sample.
