Product overview
Human total Prostate Specific Antigen ELISA kit has been re-developed. We have identified a new recombinant monoclonal antibodies to provide improved performance and consistancy. This version will be discontinued when inventory is depleted. The new version is available as ab264615.
Human Total Prostate Specific Antigen ELISA Kit (ab188388) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Total Prostate Specific Antigen protein in cell culture supernatant, cit plasma, edta plasma, hep plasma, serum, and urine. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Total Prostate Specific Antigen with 9.4 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies
- Fully validated in biological samples
- 96-wells plate breakable into 12 x 8 wells stripsA 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

Notes
Prostate specific antigen (PSA) or also commonly termed tissue Kallikrein-3 (KLK3) is a 34 kD serine protease from the Kallikrein family. PSA has very high expression in the prostate gland but expression also has been reported in the breast and saliva. PSA functions by hydrolyzing semenogelin-1 leading to the liquefaction of the seminal coagulum. This is essential for the proper release of sperm during ejaculation. The majority (>70%) of PSA is known to form a complex with alpha-1-antichymotrypsin (PSA-ACT) in the blood. The remaining PSA exists as an unbound inactive enzyme known as Free PSA. PSA is best characterized as a clinical biomarker for Prostate Cancer. During prostate cancer progression, excess amounts of PSA are leaked into the blood dramatically elevating both Total and Free PSA levels.

SimpleStep ELISA technology allows the formation of the antibody-antigen complex in one single step, reducing assay time to 90 minutes. Add samples or standards and antibody mix to wells all at once, incubate, wash, and add your final substrate. See protocol for a detailed step-by-step guide.

Mean Human male Total PSA is 424 pg/mL and values fall within expected normal reference ranges (WHO). Observed Free PSA levels were also determined using the Free Prostate Specific Antigen (free PSA) SimpleStep ELISA ab188389. Mean Human male Free PSA is 50.2 pg/mL or 11% Free PSA and values fall within expected normal reference ranges (WHO).
-
Mean Human female Total PSA is 12.5 pg/mL (50% detectable) and values fall within expected normal reference ranges (WHO). Observed Free PSA levels were also determined using the Free Prostate Specific Antigen (free PSA) SimpleStep ELISA ab188389. Mean Human female Free PSA is 12.7 pg/mL and values fall within expected normal reference ranges (WHO).
-
Mean Human Total PSA values fall within expected normal reference ranges (WHO). Observed Free PSA levels were also determined using the Free Prostate Specific Antigen (free PSA) SimpleStep ELISA ab188389. Mean Human Free PSA values fall within expected normal reference ranges (WHO). Please note: overall Total and Free PSA levels are lower due to mixed gender samples.
-
Mean Human Total PSA values are 71.7 ng/mL and fall within expected normal reference ranges (WHO). Observed Free PSA levels were also determined using the Free Prostate Specific Antigen (free PSA) SimpleStep ELISA ab188389. Mean Human Free PSA is 61.9 ng/mL and values fall within expected normal reference ranges (WHO).
-
Mean Human Total PSA is 0.54 ng/mL in normal serum and values fall within expected normal reference ranges (WHO). Observed Total PSA in each prostate cancer donor; Donor 76, 48 ng/mL; Donor 77, 120 ng/mL; Donor 78, 40 ng/mL. Observed Free PSA levels were also determined using the Free Prostate Specific Antigen (free PSA) SimpleStep ELISA ab188389. Mean Human Free PSA is 0.03 ng/mL (5% Free PSA) and values fall within expected normal reference ranges (WHO). Observed Free PSA in each prostate cancer donor; Donor 76, 9.2 ng/mL (19.2% Free PSA); Donor 77, 49 ng/mL (40.6% Free PSA); Donor 78, 7 ng/mL (17.1% Free PSA). The significant increase observed in Total and Free PSA (including % Free PSA) correlates well with the diagnosis of Prostate Cancer.
