Product overview
Human TLR2 ELISA Kit (ab227897) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of TLR2 protein in cit plasma, edta plasma, hep plasma, milk, serum, urine, and cell lysate. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human TLR2 with 17 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies
- Fully validated in biological samples
- 96-wells plate breakable into 12 x 8 wells stripsA 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

Function
Cooperates with LY96 to mediate the innate immune response to bacterial lipoproteins and other microbial cell wall components. Cooperates with TLR1 to mediate the innate immune response to bacterial lipoproteins or lipopeptides. Acts via MYD88 and TRAF6, leading to NF-kappa-B activation, cytokine secretion and the inflammatory response. May also promote apoptosis in response to lipoproteins. Recognizes mycoplasmal macrophage-activating lipopeptide-2kD (MALP-2), soluble tuberculosis factor (STF), phenol-soluble modulin (PSM) and B.burgdorferi outer surface protein A lipoprotein (OspA-L) cooperatively with TLR6. -
Tissue specificity
Highly expressed in peripheral blood leukocytes, in particular in monocytes, in bone marrow, lymph node and in spleen. Also detected in lung and in fetal liver. Levels are low in other tissues. -
Sequence similarities
Belongs to the Toll-like receptor family.
Contains 14 LRR (leucine-rich) repeats.
Contains 1 TIR domain. -
Post-translational
modificationsGlycosylation of Asn-442 is critical for secretion of the N-terminal ectodomain of TLR2. -

SimpleStep ELISA technology allows the formation of the antibody-antigen complex in one single step, reducing assay time to 90 minutes. Add samples or standards and antibody mix to wells all at once, incubate, wash, and add your final substrate. See protocol for a detailed step-by-step guide.

The concentrations of TLR2 were measured in duplicates, interpolated from the TLR2 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 50%, plasma (EDTA) 50%, plasma (heparin) 50%, and plasma (citrate) 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).

The concentrations of TLR2 were measured in duplicates, interpolated from the TLR2 standard curves and corrected for sample dilution. Undiluted samples are as follows: milk 12%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean TLR2 concentration was determined to be 41.3 ng/mL in milk.

The concentrations of TLR2 were measured in duplicate and interpolated from the TLR2 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean TLR2 concentration was determined to be 2.3 ng/mL in unstimulated THP-1 and 6.0 ng/mL in stimulated THP-1.
