Product overview
Human TFF2 ELISA kit (ab275553) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human TFF2 protein in serum, plasma, urine, and cell and tissue extract. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human TFF2 with 3.40 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
-Single-wash protocol reduces assay time to 90 minutes or less
-High sensitivity, specificity and reproducibility from superior antibodies
-Fully validated in biological samples
-96-wells plate breakable into 12 x 8 wells strips
A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

Notes
TFF2 is a member of the trefoil factor (TFF) family which contains three secreted, protease-resistant polypeptides that help to maintain gastrointestinal system integrity. Family members are 7-12 kDa in size and contain a 3-disulfide bonds. TFF1, 2, and 3 are expressed in specific areas of the digestive tract, and promote restitution, or the resealing of superficial wounds after injury. TFF2 Inhibits gastrointestinal motility and gastric acid secretion. It has been reported to function as a structural component of gastric mucus, possibly by stabilizing glycoproteins in the mucus gel through interactions with carbohydrate side chains.

SimpleStep ELISA technology allows the formation of the antibody-antigen complex in one single step, reducing assay time to 90 minutes. Add samples or standards and antibody mix to wells all at once, incubate, wash, and add your final substrate. See protocol for a detailed step-by-step guide.

The TFF2 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of TFF2 were measured in duplicates, interpolated from the TFF2 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 10%, plasma (citrate) 20%, plasma (EDTA) 20%, plasma (heparin) 20%, and pooled urine 1:800. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean TFF2 concentration was determined to be 4.53 ng/mL in serum, 2.45 ng/mL in plasma (citrate), 2.64 ng/mL in plasma (EDTA), 1.95 ng/mL in plasma (heparin), and 394.66 ng/mL in urine.

Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean TFF2 concentration was determined to be 3.78 ng/mL with a range of 0.71 – 8.45 ng/mL.
