Human SHBG ELISA Kit (ab260070) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of SHBG protein in cit plasma, edta plasma, hep plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human SHBG with 26 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. SHBG, Sex hormone-binding globulin, is a steroid binding protein used to transport dihydrotestosterone, testosterone, and 17-beta-estradiol. It is crucial in the maintenance of plasma steroid hormone levels. This kit targets the mature, full-length, protein form which lacks the N-terminal signaling peptide.

Functions as an androgen transport protein, but may also be involved in receptor mediated processes. Each dimer binds one molecule of steroid. Specific for 5-alpha-dihydrotestosterone, testosterone, and 17-beta-estradiol. Regulates the plasma metabolic clearance rate of steroid hormones by controlling their plasma concentration. -

The SHBG standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

Interpolated concentrations of native SHBG in human serum and plasma samples. Samples were pooled from healthy 20 male and 20 female volunteers. The concentrations of SHBG were measured in duplicates, interpolated from the SHBG standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 0.2%, plasma (citrate) 0.2%, plasma (heparin) 0.2% and plasma (EDTA) 0.2%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean SHBG concentration was determined to be 1466 ng/mL in serum, 1453 ng/mL in plasma (Citrate),1559 ng/mL in plasma (Heparin), and 1348 ng/mL in plasma (EDTA).
