Human PF4 ELISA Kit (CXCL4) (ab189573) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of PF4 (CXCL4) protein in cell culture supernatant, cit plasma, edta plasma, hep plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human PF4 (CXCL4) with 24.29 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
Platelet factor-4 (PF4/CXCL4) is a 70-amino acid protein that is released from the alpha-granules of activated platelets whose major physiologic role appears to be inhibiting local antithrombin III activity and promoting coagulation. PF4 (CXCL4) has a role in inflammation and wound repair by acting as a strong chemoattractant for neutrophils, monocytes, and fibroblasts.

Released during platelet aggregation. Neutralizes the anticoagulant effect of heparin because it binds more strongly to heparin than to the chondroitin-4-sulfate chains of the carrier molecule. Chemotactic for neutrophils and monocytes. Inhibits endothelial cell proliferation, the short form is a more potent inhibitor than the longer form. -

The concentrations of PF4 were measured in duplicates, interpolated from the PF4 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 2%, plasma (citrate) 0.5%, and plasma (EDTA) 4%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean PF4 concentration was determined to be 249.53 ng/mL in serum, 622.09 ng/mL in plasma (citrate), and 117.28 ng/mL in plasma (EDTA).
