Human PAPPA ELISA Kit (ab235647) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of PAPPA protein in cit plasma, edta plasma, serum, and tissue extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human PAPPA with 8.89 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Function
Metalloproteinase which specifically cleaves IGFBP-4 and IGFBP-5, resulting in release of bound IGF. Cleavage of IGFBP-4 is dramatically enhanced by the presence of IGF, whereas cleavage of IGFBP-5 is slightly inhibited by the presence of IGF. -
Tissue specificity
High levels in placenta and pregnancy serum. In placenta, expressed in X cells in septa and anchoring villi, and in syncytiotrophoblasts in the chorionic villi. Lower levels are found in a variety of other tissues including kidney, myometrium, endometrium, ovaries, breast, prostate, bone marrow, colon, fibroblasts and osteoblasts. -

The concentrations of PAPPA were measured in duplicates, interpolated from the PAPPA standard curves and corrected for sample dilution. Undiluted samples are as follows: pregnant serum 1:500 and pregnant serum pre-eclampsia 1:500. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean PAPPA concentration was determined to be 1669.24 ng/mL in pregnant serum, and 2393.50 ng/mL in pregnant serum pre-eclampsia.
