  • Product overview

Human LRP1 ELISA kit (ab278128) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human LRP1 protein in human serum, plasma-citrate, cell culture supernatant, cell and tissue extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human LRP1 with 30.6 pg/mL sensitivity.

SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:

-Single-wash protocol reduces assay time to 90 minutes or less
-High sensitivity, specificity and reproducibility from superior antibodies
            -Fully validated in biological samples
            -96-wells plate breakable into 12 x 8 wells strips

A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

  • Notes

Prolow-density lipoprotein receptor-related protein 1 (LRP1) is an endocytic receptor involved in endocytosis and in phagocytosis of apoptotic cells.  LRP1 is widely expressed on many cell types including neurons, adipocytes, hepatocytes, vascular smooth muscle cells, fibroblasts, keratinocytes, macrophages, and megakaryocytes.  Human LRP1 is an N-glycosylated and sialylated molecule that is cleaved in the Golgi apparatus to produce an 85 kDa transmembrane beta chain and a 515 kDa alpha chain that associates noncovalently with the beta chain but does not itself cross the membrane.  The provided protein standard for ab278128 is a fragment of the extracellular domain.

  • Function

Endocytic receptor involved in endocytosis and in phagocytosis of apoptotic cells. Required for early embryonic development. Involved in cellular lipid homeostasis. Involved in the plasma clearance of chylomicron remnants and activated LRPAP1 (alpha 2-macroglobulin), as well as the local metabolism of complexes between plasminogen activators and their endogenous inhibitors. May modulate cellular events, such as APP metabolism, kinase-dependent intracellular signaling, neuronal calcium signaling as well as neurotransmission.
Functions as a receptor for Pseudomonas aeruginosa exotoxin A.
  • Tissue specificity

Most abundant in liver, brain and lung.
  • Sequence similarities

    Belongs to the LDLR family.
    Contains 22 EGF-like domains.
    Contains 31 LDL-receptor class A domains.
    Contains 34 LDL-receptor class B repeats.
  • Post-translational
    modifications

Cleaved into a 85 kDa membrane-spanning subunit (LRP-85) and a 515 kDa large extracellular domain (LRP-515) that remains non-covalently associated. Gamma-secretase-dependent cleavage of LRP-85 releases the intracellular domain from the membrane.
    The N-terminus is blocked.
Phosphorylated on serine and threonine residues.
Phosphorylated on tyrosine residues upon stimulation with PDGF. Tyrosine phosphorylation promotes interaction with SHC1.
  • Cellular localization

Cytoplasm. Nucleus. After cleavage, the intracellular domain (LRPICD) is detected both in the cytoplasm and in the nucleus and Cell membrane. Membrane, coated pit.

SimpleStep ELISA technology allows the formation of the antibody-antigen complex in one single step, reducing assay time to 90 minutes. Add samples or standards and antibody mix to wells all at once, incubate, wash, and add your final substrate. See protocol for a detailed step-by-step guide.
