Human IRAK-1 ELISA Kit (ab219630) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of IRAK-1 protein in cell culture extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human IRAK-1 with 22 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
IRAK-1 is a serine/threonine-protein kinase involved in the innate immune response against pathogens. IRAK-1’s association with MYC88 after TLR activation leads to its phosphorylation by IRAK-4. IRAK-1 phosphorylation eventually results in the transcriptional activation of type 1 IFN genes.

Function
Binds to the IL-1 type I receptor following IL-1 engagement, triggering intracellular signaling cascades leading to transcriptional up-regulation and mRNA stabilization. Isoform 1 binds rapidly but is then degraded allowing isoform 2 to mediate a slower, more sustained response to the cytokine. Isoform 2 is inactive suggesting that the kinase activity of this enzyme is not required for IL-1 signaling. Once phosphorylated, IRAK1 recruits the adapter protein PELI1. -
Tissue specificity
Isoform 1 and isoform 2 are ubiquitously expressed in all tissues examined, with isoform 1 being more strongly expressed than isoform 2. -

The concentrations of IRAK-1 were measured in duplicate and interpolated from the IRAK-1 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IRAK-1 concentration was determined to be 8178 pg/mL in MCF7 extracts, 3768 pg/mL in HEK93T extracts, and 3927 pg/mL in K562 extracts.
