Human iNOS ELISA Kit (ab253217) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of iNOS protein in cell lysate. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human iNOS with 18 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
iNOS, also known as NOS2, catalyzes the production of nitric oxide from L-arginine, which is used in various cell signaling events. iNOS is an inducible member of the NOS family and is upregulated as a host-defense mechanism during proinflammatory cytokine activity seen with tumorigenesis and parasitic infection. iNOS activity is a key cause of septic shock.

Function
Produces nitric oxide (NO) which is a messenger molecule with diverse functions throughout the body. In macrophages, NO mediates tumoricidal and bactericidal actions. Also has nitrosylase activity and mediates cysteine S-nitrosylation of cytoplasmic target proteins such COX2. -
Tissue specificity
Expressed in the liver, retina, bone cells and airway epithelial cells of the lung. Not expressed in the platelets. -

The iNOS standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of iNOS were measured in duplicate and interpolated from the iNOS standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean iNOS concentration was determined to be 613.7 pg/mL in Caco-2 cell extract samples.
