Product overview
Human IL-6 Receptor (Interleukin-6 Receptor) ELISA kit (ab181424) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of IL-6 Receptor protein in human serum, plasma and cell culture supernatant. It uses our proprietary SimpleStep ELISA® technology. Quantitate human IL-6 Receptor with 46 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
-Single-wash protocol reduces assay time to 90 minutes or less
-High sensitivity, specificity and reproducibility from superior antibodies
-Fully validated in biological samples
-96-wells plate breakable into 12 x 8 wells strips
A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpeStep ELISA® kits. -
Notes
Interleukin-6 Receptor (IL-6R, IL-6 Receptor), also known as Interleukin-6 Receptor subunit alpha and membrane glycoprotein 80, is part of a receptor that binds Interleukin-6 (IL-6). IL-6 Receptor weakly binds IL-6 with a low affinity, and then is presented to gp130 to facilitate transition into a high affinity signaling competent hexamer. Activation may lead to regulation of the immune response, acute-phase reactions and hematopoiesis. The production of IL-6 Receptor has been implicated in the pathogenesis of many diseases, including multiple myeloma, auto immune diseases and prostate cancer.IL-6 Receptor has two isoforms. The first is a transmembrane protein in which the N-terminal extracellular portion can be released by proteolytic cleavage. The second isoform is a secreted form that lacks the transmembrane and intracellular portions. This assay is directed at the extracellular portion of IL-6 Receptor and detects both the soluble and membrane bound forms.

Function
Part of the receptor for interleukin 6. Binds to IL6 with low affinity, but does not transduce a signal. Signal activation necessitate an association with IL6ST. Activation may lead to the regulation of the immune response, acute-phase reactions and hematopoiesis.
Low concentration of a soluble form of IL6 receptor acts as an agonist of IL6 activity. -
Tissue specificity
Isoform 2 is expressed in peripheral blood mononuclear cells and weakly found in urine and serum. -
Sequence similarities
Belongs to the type I cytokine receptor family. Type 3 subfamily.
Contains 1 fibronectin type-III domain.
Contains 1 Ig-like C2-type (immunoglobulin-like) domain. -
Domain
The two fibronectin type-III-like domains, contained in the N-terminal part, form together a cytokine-binding domain.
The WSXWS motif appears to be necessary for proper protein folding and thereby efficient intracellular transport and cell-surface receptor binding. -
Post-translational
modificationsA short soluble form may also be released from the membrane by proteolysis. -
Cellular localization
Secreted and Basolateral cell membrane. - Information by UniProt

SimpleStep ELISA technology allows the formation of the antibody-antigen complex in one single step, reducing assay time to 90 minutes. Add samples or standards and antibody mix to wells all at once, incubate, wash, and add your final substrate. See protocol for a detailed step-by-step guide.

Background-subtracted data values (mean +/- SD) are graphed.
-
The concentrations of IL-6 Receptor were interpolated from the IL-6 Receptor standard curve and corrected for sample dilution.
-
The concentrations of IL-6 Receptor were interpolated the IL-6 Receptor standard curve and corrected for sample dilution. The mean IL-6 Receptor concentration was determined to be 27,796 pg/mL in plasma (EDTA) and 25,731 pg/mL in serum.
-
Serum from 10 apparently healthy male donors was measured in duplicate for IL-6 Receptor. The mean IL-6 Receptor concentration was determined to be 24,806 pg/mL with a range of 17,960 – 35,807 pg/mL.
