Product overview
Human IGFBP1 ELISA Kit (ab233618) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of IGFBP1 protein in cit plasma, edta plasma, hep plasma, milk, serum, tissue homogenate, urine, cell culture supernatant, and cell culture extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human IGFBP1 with 11.6 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies
- Fully validated in biological samples
- 96-wells plate breakable into 12 x 8 wells stripsA 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

Function
IGF-binding proteins prolong the half-life of the IGFs and have been shown to either inhibit or stimulate the growth promoting effects of the IGFs on cell culture. They alter the interaction of IGFs with their cell surface receptors. Promotes cell migration. -
Sequence similarities
Contains 1 IGFBP N-terminal domain.
Contains 1 thyroglobulin type-1 domain. -
Post-translational
modificationsPhosphorylated; probably by casein kinase II. Alters the affinity of the protein for IGFs. -
Cellular localization
Secreted. - Information by UniProt

SimpleStep ELISA technology allows the formation of the antibody-antigen complex in one single step, reducing assay time to 90 minutes. Add samples or standards and antibody mix to wells all at once, incubate, wash, and add your final substrate. See protocol for a detailed step-by-step guide.

Standard curve comparison between Human IGFBP1 SimpleStep ELISA® kit and traditional ELISA kit from leading competitor. SimpleStep ELISA kit shows increased sensitivity.
-
The IGFBP1 standard curve was prepared as described in Section 10.
-
-
The concentrations of IGFBP1 were measured in duplicates, interpolated from the IGFBP1 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 2.5%, plasma (citrate) 5%, plasma (heparin) 5%, and plasma (EDTA) 2.5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IGFBP1 concentration was determined to be 22 ng/mL in serum, 16 ng/mL in plasma (citrate), 21 ng/ml in plasma (heparin), and 37 ng/ml in plasma (EDTA).

The concentrations of IGFBP1 were measured in duplicates, interpolated from the IGFBP1 standard curves and corrected for sample dilution. Undiluted samples are as follows: milk 5%, A549 supernatant 100%, and HepG2 supernatant 0.5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IGFBP1 concentration was determined to be 12 ng/ml in milk, 0.5 ng/ml in A549 supernatant, and 172 ng/mL in HepG2 supernatant.
-
. Interpolated concentrations of native IGFBP1 in human HepG2, U-87 MG, and liver homogenate extract samples based on a 4 µg/mL, 500 µg/ml, and 500 µg/mL extract load respectively
The concentrations of IGFBP1 were measured in duplicate and interpolated from the IGFBP1 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IGFBP1 concentration was determined to be 580 pg/mL in HepG2 cell extract, 457 pg/ml in U-87 MG cell extract, and 739 pg/mL in liver tissue homogenate extract samples.
-
Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IGFBP1 concentration was determined to be 29.1 ng/mL with a range of 1.5 – 117.3 ng/mL.
