Product overview
Human ICAM3 ELISA kit (ab275097) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human ICAM3 protein in serum, plasma and cell culture supernatant. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human ICAM3 with 0.036 ng/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
-Single-wash protocol reduces assay time to 90 minutes or less
-High sensitivity, specificity and reproducibility from superior antibodies
-Fully validated in biological samples
-96-wells plate breakable into 12 x 8 wells strips
A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

Notes
Intracellular adhesion molecule 3 (ICAM-3), also known as CD50, is a protein encoded by the gene ICAM3. ICAM3 belongs to the ICAM family of proteins, where all ICAM proteins are type I transmembrane glycoproteins, contain several immunoglobulin-like c2 domains, and bind to leukocyte adhesion protein (LFA-1). In addition, ICAM-3 is a ligand for integrin alpha-D/beta-2, where it contributes to apoptotic neutrophil phagocytosis by macrophages.

Function
ICAM proteins are ligands for the leukocyte adhesion protein LFA-1 (integrin alpha-L/beta-2). ICAM3 is also a ligand for integrin alpha-D/beta-2. -
Sequence similarities
Belongs to the immunoglobulin superfamily. ICAM family.
Contains 5 Ig-like C2-type (immunoglobulin-like) domains. -
Post-translational
modificationsUpon stimulation by a physiologic stimuli becomes rapidly and transiently phosphorylated on serine residues.
N-glycosylated; glycans consist of a mixture of tri- and tetra-antennary complex-type chains and high-mannose chains. -

SimpleStep ELISA technology allows the formation of the antibody-antigen complex in one single step, reducing assay time to 90 minutes. Add samples or standards and antibody mix to wells all at once, incubate, wash, and add your final substrate. See protocol for a detailed step-by-step guide.

The ICAM3 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.
-
The concentrations of ICAM3 were measured in duplicates, interpolated from the target standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 12.5%, plasma (EDTA) 12.5%, plasma (citrate) 12.5%, and plasma (heparin) 12.5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean target concentration was determined to be 124.4 ng/mL in serum, 87.3 ng/mL in plasma (EDTA), 96.7 ng/mL in and plasma (citrate), 105 ng/mL in plasma (heparin).
