Product overview
Human HC-II ELISA Kit (ab277402) is an in-vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human HC-II in serum, plasma and cell culture supernatants.
This assay employs an antibody specific for Human HC-II coated on a 96-well plate. Standards and samples are pipetted into the wells and Human HC-II present in a sample is bound to the wells by the immobilized antibody. The wells are washed, and biotinylated anti-Human HC-II antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Human HC-II bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Function
Thrombin inhibitor activated by the glycosaminoglycans, heparin or dermatan sulfate. In the presence of the latter, HC-II becomes the predominant thrombin inhibitor in place of antithrombin III (AT-III). Also inhibits chymotrypsin, but in a glycosaminoglycan-independent manner.
Peptides at the N-terminal of HC-II have chemotactic activity for both monocytes and neutrophils. -
Tissue specificity
Expressed predominantly in liver. Also present in plasma. -
Involvement in disease
Thrombophilia due to heparin cofactor 2 deficiency -
Sequence similarities
Belongs to the serpin family. -
Domain
The N-terminal acidic repeat region mediates, in part, the glycosaminoglycan-accelerated thrombin inhibition. -
Post-translational
modificationsPhosphorylation sites are present in the extracellular medium. - Information by UniProt
