Human Glypican 1 ELISA Kit (GPC1) (ab270217) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Glypican 1 (GPC1) protein in cell lysate, edta plasma, serum, urine, cell culture extracts, tissue extracts, and cell culture supernatant. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Glypican 1 (GPC1) with 16.16 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
Human Glypican 1 (GPC1) is synthesized as a 558 amino acid (aa) precursor that contains a 23 aa signal sequence, a 507 aa mature segment, and a 28 aa C-terminal prosegment. The standard protein in this product is the mature form. Members of the glypican-related integral membrane proteoglycan family (GRIPS) contain a core protein anchored to the cytoplasmic membrane via a glycosyl phosphatidylinositol linkage. GPC1 may play a role in the control of cell division and growth regulation. Human GPC1 shares 89%, 89%, and 87% sequence homology with mouse, rat, and bovine, respectively.

The concentrations of Glypican 1 were measured in duplicates, interpolated from the Glypican 1 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 25%, plasma (citrate) 50%, plasma (EDTA) 50%, and plasma (heparin) 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Glypican 1 concentration was determined to be 11.98 ng/mL in serum, 9.90 ng/mL in plasma (citrate) and 10.44 ng/mL in plasma (EDTA), and 9.93 ng/mL in plasma (Heparin).

Interpolated concentrations of native Glypican 1 in human cell culture supernatant, and urine samples.
The concentrations of Glypican 1 were measured in duplicates, interpolated from the Glypican 1 standard curves and corrected for sample dilution. Undiluted samples are as follows: MCF7 supernatant 25%, HepG2 supernatant 50%, and urine 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Glypican 1 concentration was determined to be 10.14 ng/mL in MCF7 supernatant, 4.5 ng/mL in HepG2 supernatant and 7.87 ng/mL in urine.
