Human Ficolin-2 ELISA kit (ab275554) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human Ficolin-2 protein in serum, plasma and urine. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Ficolin-2 with 8.35 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
Ficolin-2, initially identified as L-ficolin, in human is encoded by the FCN2 gene. Ficolin-2 belongs to the ficolin family of proteins and is characterized by the presence of a leader peptide, a short N-terminal segment, followed by a collagen-like region, and a C-terminal fibrinogen-like domain. Ficolin-2 is predominantly expressed in the liver. Studies have reported it to have roles in carbohydrate binding and opsonic activities. Serum concentration of ficolin-2 varies considerably in healthy individuals. FCN2 promoter polymorphisms were associated with marked changes in the ficolin-2 serum concentration.

The Ficolin-2 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of Ficolin-2 were measured in duplicates, interpolated from the Ficolin-2 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 1:200, plasma (citrate) 1:200, plasma (EDTA) 1:400, and plasma (heparin) 1:200. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Ficolin-2 concentration was determined to be 481.67 ng/mL in serum, 508.04 ng/mL in plasma (citrate), 443.89 ng/mL in plasma (EDTA), and 500.03 ng/mL in plasma (heparin).

Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Ficolin-2 concentration was determined to be 403.80 ng/mL with a range of 138.63 – 755.53 ng/mL.
