Human Fetuin-A ELISA Kit (ab269372) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Fetuin-A protein in cell culture supernatant, edta plasma, serum, and cit plasma. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Fetuin-A with 116.24 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
Fetuin A is a highly abundant protein secreted by the liver that is capable of binding a wide variety of ligands, similar to serum albumin. A member of the cystatin family, Fetuin A plays a vital role in preventing tissue calcification and is also involved in fatty acid transport, insulin regulation and anti-inflammatory responses. Fetuin A is encoded by a single gene on chromosome 3, contains two cystatin domains, and exists as a heterodimer in its posttranslational form. Fetuin A is more abundant than serum albumin in fetal serum.

The Fetuin-A standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of Fetuin-A were measured in duplicates, interpolated from the Fetuin-A standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 1: 20,000, plasma (citrate) 1: 20,000, plasma (EDTA) 1: 20,000, and HepG2 supernatant 1: 600. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Fetuin-A concentration was determined to be 194.15 µg/mL in serum, 164.51 µg/mL in plasma (citrate), 172.44 µg/ml in plasma (EDTA), and 3.85 µg/mL HepG2 supernatant.

Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Fetuin-A concentration was determined to be 170.03 µg/mL with a range of 136.89 – 246.74 µg/mL.
