Product overview
Human CD85d ELISA Kit (LILRB2) (ab269551) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of CD85d (LILRB2) protein in edta plasma, hep plasma, serum, and cit plasma. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human CD85d (LILRB2) with 15.9 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies
- Fully validated in biological samples
- 96-wells plate breakable into 12 x 8 wells stripsA 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

Notes
CD85d (LILRB2), Leukocyte immunoglobulin-like receptor subfamily B member 2, is a key protein in the regulation of the immune response where it plays a pivotal as a negative regulator. It is primarily found on immune cells where it interacts with MHC domains on antigen presenting cells. Given its important role in immunosuppression, it is currently under investigation as a drug-targetable protein for a host of disease states. The antibodies for this kit were raised against a recombinant protein fragment targeting the extracellular domain of this receptor. There is little homology between mouse and rat for this fragment, so reactivity is not likely.

Function
Receptor for class I MHC antigens. Recognizes a broad spectrum of HLA-A, HLA-B, HLA-C and HLA-G alleles. Involved in the down-regulation of the immune response and the development of tolerance. Competes with CD8A for binding to class I MHC antigens. Inhibits FCGR1A-mediated phosphorylation of cellular proteins and mobilization of intracellular calcium ions. -
Tissue specificity
Expressed on monocytes and B-cells, and at lower levels on dendritic cells. Detected at low levels in natural killer (NK) cells. -
Sequence similarities
Contains 4 Ig-like C2-type (immunoglobulin-like) domains. -
Domain
Contains 3 copies of a cytoplasmic motif that is referred to as the immunoreceptor tyrosine-based inhibitor motif (ITIM). This motif is involved in modulation of cellular responses. The phosphorylated ITIM motif can bind the SH2 domain of several SH2-containing phosphatases. -
Post-translational
modificationsPhosphorylated on tyrosine residues. Dephosphorylated by PTPN6. -

SimpleStep ELISA technology allows the formation of the antibody-antigen complex in one single step, reducing assay time to 90 minutes. Add samples or standards and antibody mix to wells all at once, incubate, wash, and add your final substrate. See protocol for a detailed step-by-step guide.

The CD85d standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.
-
The concentrations of CD85d were measured in duplicates, interpolated from the CD85d standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 30%, plasma (EDTA) 30%, plasma (citrate) 30%, and plasma (heparin) 30%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean CD85d concentration was determined to be 8.85 ng/mL in serum, 6.75 ng/mL in plasma (EDTA), 8.82 ng/mL in plasma (citrate), and 7.72 ng/mL in plasma (Heparin).
