Human CCL14 ELISA kit (ab272201) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of CCL14 protein in serum, plasma and cell culture supernatant. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human CCL14 with 4.4 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
C-C motif chemokine 14 (CCL14), also known as HCC-1 and NCC-2, is a cytokine encoded by the gene CCL14. Mature secreted CCL14 is a 74- amino acid containing active glycoprotein and is constitutively expressed by multiple tissues and has been found at high concentrations in plasma.
CCl14 enhances the proliferation of CD34 myeloid progenitor cells and has weak activities on human monocytes through receptors that also recognize MIP-1 alpha. CCL14 induces intracellular Ca2+ changes and enzyme release but does not induce chemotaxis in monocytes.

Function
Has weak activities on human monocytes and acts via receptors that also recognize MIP-1 alpha. It induced intracellular Ca(2+) changes and enzyme release, but no chemotaxis, at concentrations of 100-1,000 nM, and was inactive on T-lymphocytes, neutrophils, and eosinophil leukocytes. Enhances the proliferation of CD34 myeloid progenitor cells. The processed form HCC-1(9-74) is a chemotactic factor that attracts monocytes eosinophils, and T-cells and is a ligand for CCR1, CCR3 and CCR5. -

Expressed constitutively in several normal tissues: spleen, liver, skeletal and heart muscle, gut, and bone marrow, present at high concentrations (1-80 nM) in plasma. -

The CCL14 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of CCL14 were measured in duplicates, interpolated from the target standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 1:200, plasma (EDTA) 1:200, plasma (citrate) 1:200, plasma (heparin) 1:400. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean target concentration was determined to be 59.0 ng/mL in serum, 43.1 ng/mL in plasma (EDTA), 48.8 ng/mL in and plasma (citrate), and 98.9 ng/mL in plasma (heparin).
