Human Caspase 4 ELISA kit (ab275098) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human Caspase 4 protein in cell culture and cell and tissue extract. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Caspase 4 with 6.7 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
Human Caspase-4 also known as ICE and Ced-3 homolog 2, Mih1, and protease TX, is a protein in the family of cysteine proteases called caspases and is encoded by the gene CASP4. CASP4 is expressed as an inactive proenzyme comprised of a pro-domain and a large and small protease subunit. CASP4 is activated by cleavage of an internal Aspartic residue, thus generates a heterodimeric enzyme consisting of the two subunits which is capable of cleaving and activating its own precursor protein. Overexpression of CASP4 results in apoptosis. The antibodies used in this kit were immunized against regions in the large subunit (subunit 1).

The Caspase 4 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of Caspase 4 were measured in duplicate and interpolated from the Caspase 4 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).
