Abcam’s anti-ENA Screen Human in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the accurate semi-quantitative measurement of IgG class antibodies against extractable nuclear antigens: Sm (Smith), RNP/Sm, Scl-70, SS-A (Ro) (52kDa and 60kDa), SS-B (La), Jo-1, in Human serum or plasma.
A 96-well plate has been precoated with ENA antigens to bind cognate antibodies. Controls, standards or test samples are added to the wells and incubated. Following washing, a horseradish peroxidase (HRP) labelled anti-Human IgG conjugate is added to the wells, which binds to the immobilized ENA-specific antibodies. TMB is then catalyzed by the HRP to produce a blue color product that changes to yellow after adding an acidic stop solution. The density of yellow coloration is directly proportional to the amount of ENA IgG sample captured in plate.

ENA is a grouping of antibodies often used to screen for mixed connective tissue disease (MCTD), Sjögren's syndrome and systemic lupus erythematosus and commonly is composed of four tests.
