Human ACY-1 ELISA Kit (ab277463) is an in-vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human ACY-1 in serum, plasma (collect plasma using EDTA as an anticoagulant. Heparin and Citrate are not recommended as anticoagulants for use in this assay) and cell culture supernatants.
This assay employs an antibody specific for Human ACY-1 coated on a 96-well plate. Standards and samples are pipetted into the wells and Human ACY-1 present in a sample is bound to the wells by the immobilized antibody. The wells are washed, and biotinylated anti-Human ACY-1 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Human ACY-1 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Involvement in disease
Defects in ACY1 are the cause of aminoacylase-1 deficiency (ACY1D) [MIM:609924]. ACY1D results in a metabolic disorder manifesting with encephalopathy, unspecific psychomotor delay, psychomotor delay with atrophy of the vermis and syringomyelia, marked muscular hypotonia or normal clinical features. Epileptic seizures are a frequent feature. All affected individuals exhibit markedly increased urinary excretion of several N-acetylated amino acids. -
