Cathepsin H Activity Assay Kit (Fluorometric) (ab65305) is a fluorescence-based assay that utilizes the preferred cathepsin-B substrate Arginine labeled with AFC (amino-4-trifluoromethyl coumarin). Cell lysates or other samples that contain cathepsin-B will cleave the synthetic substrate R-AFC to release free AFC. The released AFC can easily be quantified using a fluorometer or fluorescence plate reader.The released AFC can easily be quantified using a fluorometer or fluorescence plate reader. The Cathepsin H assay is simple, straightforward, and can be adapted to 96-well plate assays. Assay conditions have been optimized to obtain the maximal activity.

Notes
Apoptosis can be mediated by mechanisms other than the traditional caspase-mediated cleavage cascade. There is growing recognition that alternative proteolytic enzymes such as the lysosomal cathepsin proteases may initiate or propagate proapoptotic signals. Cathepsins are lysosomal enzymes that are also used as sensitive markers in various toxicological investigations.

Relevance
The cathepsin family of proteolytic enzymes contains several diverse classes of proteases. The cysteine protease class comprises cathepsins B, L, H, K, S, and O. The aspartyl protease class is composed of cathepsins D and E. Cathepsin G is in the serine protease class. Most cathepsins are lysosomal and each is involved in various cellular events such as peptide biosynthesis and protein degradation. The propeptide region of cathepsin H is highly homologous to other cathepsins. This region is thought to play a role in the processing of cysteine proteases. Cathepsin H is thought to be involved in several pathological conditions, including neuro degenerative disorders and cancer. -
