Abcam's Caspase 5 Assay Kit (Fluorometric) provides a simple and convenient means for assaying the activity of caspases that recognize the sequence WEHD. The assay is based on detection of cleavage of substrate WEHD-AFC (AFC: 7-amino-4-trifluoromethyl coumarin). WEHD-AFC emits blue light (λmax = 400 nm); upon cleavage of the substrate by caspase 5 or related caspases, free AFC emits a yellow-green fluorescence (λmax = 505 nm), which can be quantified using a fluorometer or a fluorecence microtiter plate reader.

Relevance
Caspase 5 belongs to the Ced3/ICE family of cysteine proteases. Caspase 5 exists as an inactive proenzyme which undergoes proteolytic processing at conserved aspartic residues, leading to p20 and p10 subunits that dimerize to form the active enzyme. Sequential activation of caspases plays a central role in the execution-phase of cell apoptosis. -
