Product overview
Osteopontin in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of Osteopontin protein in dog plasma, serum and urine.

Notes
In this assay the Osteopontin present in samples reacts with the anti-Osteopontin antibodies which have been adsorbed to the surface of polystyrene microtitre wells. After the removal of unbound proteins by washing, the Detection Antibody, biotin conjugated anti-Osteopontin, is added and complexes are formed. Following a wash step, horseradish peroxidase (HRP) conjugated Streptavidin is added and complexes are formed. After another washing step, the complexes are assayed by the addition of a chromogenic substrate, 3,3’,5,5’-tetramethylbenzidine (TMB). The quantity of bound enzyme varies directly with the concentration of Osteopontin in the sample tested; thus, the absorbance, at 450 nm, is a measure of the concentration of Osteopontin in the test sample. The quantity of Osteopontin in the test sample can be interpolated from the standard curve constructed from the standards, and corrected for sample dilution.

Function
Binds tightly to hydroxyapatite. Appears to form an integral part of the mineralized matrix. Probably important to cell-matrix interaction.
Acts as a cytokine involved in enhancing production of interferon-gamma and interleukin-12 and reducing production of interleukin-10 and is essential in the pathway that leads to type I immunity. -
Tissue specificity
Bone. Found in plasma. -
Sequence similarities
Belongs to the osteopontin family. -
Post-translational
modificationsExtensively phosphorylated on clustered serine residues.
N- and O-glycosylated.
Phosphorylation sites are present in the extracelllular medium. -
Cellular localization
Secreted. - Information by UniProt
