Chromogenic beta-galactosidase substrate -
Biological description
Chromogenic substrate for β-galactosidase, used to determine the presence or absence of a cloned DNA insert in bacteria growing on agar plates. Designed as an alternative to X-Gal (ab144388) in blue-white selection of recombinant bacterial colonies with the lac+ phenotype, producing a darker blue color.

General notes
Instructions for use: Add 5-Bromo-3-indoxyl- beta-D-galacto-pyranoside (BB1182) as concentrated stock solution (solvent DMSO or DMF) to autoclaved media
Recommended working concentration 0.1-0.3 mM (1-3 mL of 100 mM stock solution per L)
For best results also add 0.1-1 mM of the inducer isopropyl-beta-D-thiogalactopyrano-side (IPTG) to growth media

Handling
Wherever possible, you should prepare and use solutions on the same day. However, if you need to make up stock solutions in advance, we recommend that you store the solution as aliquots in tightly sealed vials at -20°C. Generally, these will be useable for up to one month. Before use, and prior to opening the vial we recommend that you allow your product to equilibrate to room temperature for at least 1 hour.
