Purified from rabbit serum by sequential epitope-specific chromatography. The antibody has been negatively preadsorbed using a non-phosphopeptide corresponding to the site of phosphorylation to remove antibody that is reactive with non-phosphorylated tau. The final product is generated by affinity chromatography using a tau-derived peptide that is phosphorylated at serine 409. -

Recombinant human tau was (a) untreated, (b) phosphorylated in vitro by casein kinase II, and (c) phosphorylated in vitro by the catalytic subunit of protein kinase A. Protein was resolved by SDS-PAGE on a 4-20% Tris glycine gel and transferred to PVDF. Membranes were incubated with 1/1000 dilution of pan tau antibody or 0.5
µ g/mL anti-tau [pS409]. After washing, membranes were incubated with goat F(ab’)2 antirabbit IgG or anti-mouse IgG alkaline phosphatase and bands were detected using the Tropix WesternStar detection method. The data show that the anti-tau pS409 antibody only recognizes recombinant tau protein phosphorylated by the catalytic subunit of protein kinase A, demonstrating the specificity of this antibody.
