ChIP- qPCR analysis of STAT6 was performed with 3 μg/mL of ab277765 on sheared chromatin from 2 million HeLa (Human epithelial cell line from cervix adenocarcinoma) cells treated with IFN-gamma (50 ng/mL) for 1h using the MAGnify Chromatin Immunoprecipitation System. Normal rabbit IgG (3 μg/mL) was used as a negative IP control. The purified DNA from each ChIP sample was analyzed by StepOnePlus Real-Time PCR System with primers for the promoter of active AP-1 and COX2 gene, used as positive control targets, and the inactive SAT2 satellite repeat, used as negative control target. Data is presented as fold enrichment of the antibody signal versus the negative control IgG using the comparative CT method.
