Target

  • Function

Met- and Leu-enkephalins compete with and mimic the effects of opiate drugs. They play a role in a number of physiologic functions, including pain perception and responses to stress. PENK(114-133) and PENK(237-258) increase glutamate release in the striatum. PENK(114-133) decreases GABA concentration in the striatum.
  • Sequence similarities

Belongs to the opioid neuropeptide precursor family.
  • Post-translational
    modifications

The N-terminal domain contains 6 conserved cysteines thought to be involved in disulfide bonding and/or processing.
  • Cellular localization

    Secreted.
  • Target information above from: UniProt accession P01210 The UniProt Consortium
The Universal Protein Resource (UniProt) in 2010
    Nucleic Acids Res. 38:D142-D148 (2010) .

    Western blot - Anti-Enkephalin/ENK antibody (ab85798)
    Anti-Enkephalin/ENK antibody (ab85798) at 1 µg/ml + Human testis tissue lysate - total protein (ab30257) at 10 µg

    Secondary
Goat polyclonal Secondary Antibody to Rabbit IgG - H&L (HRP), pre-adsorbed at 1/5000 dilution

    Developed using the ECL technique.

    Performed under reducing conditions.

    Predicted band size: 30 kDa
    Observed band size: 28 kDa
why is the actual band size different from the predicted?

    Exposure time: 30 seconds

The band observed at 28 kDa could potentially be a cleaved form of Enkephalin due to the presence of a 24 amino acid signal peptide.

ICC/IF image of ab85978 stained HeLa cells. The cells were 4% PFA fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab85798, 5µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM. This antibody also gave a positive result in 4% PFA fixed (10 min) Hek293, HepG2 and MCF7 cells at 5µg/ml, and in 100% methanol fixed (5 min) HeLa, Hek293, HepG2 and MCF7 cells at 5µg/ml.
