General notes
Neisseria gonorrhoeae is grown on permissive agar plates in a sterile class 2 containment hood. The plates are incubated at 37°C in the presence of 5% CO2 for 3 days. When optimal growth of colonies is seen, biomass is scraped off the plate and resuspended in sterile PBS.
A saline solution of N. gonorrhoeae is disrupted with 0.5 micron diameter glass beads to break open cells and expose more antigens. The product is then inactivated by heating to 56 °C for 3 hours and 0.09% sodium azide is added as a preservative.
Inactivation: 56 degrees for 3 hours, confirmed inactive by no visible colonies on permissive plates after 5 days.
Vortex/sonicate before use to ensure a homogenous solution.
This antigen is derived from a pathogenic organism, and may be involved in a disease process, consequently exposure may have adverse health effects.
