Product overview
Mouse SP-D ELISA Kit (ab240683) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of SP-D protein in cit plasma, edta plasma, hep plasma, serum, tissue extracts, and cell culture supernatant. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse SP-D with 2.16 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies
- Fully validated in biological samples
- 96-wells plate breakable into 12 x 8 wells stripsA 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

Research areas
-
Function
Contributes to the lung's defense against inhaled microorganisms. May participate in the extracellular reorganization or turnover of pulmonary surfactant. Binds strongly maltose residues and to a lesser extent other alpha-glucosyl moieties. -
Sequence similarities
Belongs to the SFTPD family.
Contains 1 C-type lectin domain.
Contains 1 collagen-like domain. -
Post-translational
modificationsThe N-terminus is blocked.
Hydroxylation on proline residues within the sequence motif, GXPG, is most likely to be 4-hydroxy as this fits the requirement for 4-hydroxylation in vertebrates. -
Cellular localization
Secreted > extracellular space > extracellular matrix. Secreted > extracellular space > surface film. - Information by UniProt

SimpleStep ELISA technology allows the formation of the antibody-antigen complex in one single step, reducing assay time to 90 minutes. Add samples or standards and antibody mix to wells all at once, incubate, wash, and add your final substrate. See protocol for a detailed step-by-step guide.

The SP-D standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of SP-D were measured in duplicates, interpolated from the SP-D standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 20%, plasma (citrate) 15%, plasma (EDTA) 30%, and plasma (heparin) 30%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean SP-D concentration was determined to be 4172.67 pg/mL in serum, 5466.91 pg/mL in plasma (citrate), 3129.78 pg/mL in plasma (EDTA), and 2563.11 pg/mL in plasma (heparin).

The concentrations of SP-D were measured in duplicates, interpolated from the SP-D standard curves and corrected for sample dilution. Undiluted samples are as follows: LPS treated lung supernatant 1%, and untreated lung supernatant 2.5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean SP-D concentration was determined to be 82.93 ng/mL in LPS treated lung supernatant and 26.10 ng/mL in untreated lung supernatant.
