Product overview
Mouse SLAM ELISA Kit is designed for the quantitative determination of SLAM in cell culture supernatants, plasma and serum samples.
Δ Note: Collect plasma using citrate and heparin as an anticoagulant. EDTA is not recommended as an anticoagulant for use in this assay due to its chelating properties.
This assay employs an antibody specific for mouse SLAM coated on a 96-well plate. Standards and samples are pipetted into the wells and SLAM present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-mouse SLAM antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of SLAM bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Function
High-affinity self-ligand important in bidirectional T-cell to B-cell stimulation. SLAM-induced signal-transduction events in T-lymphocytes are different from those in B-cells. Two modes of SLAM signaling are likely to exist: one in which the inhibitor SH2D1A acts as a negative regulator and another in which protein-tyrosine phosphatase 2C (PTPN11)-dependent signal transduction operates. -
Tissue specificity
Constitutively expressed on peripheral blood memory T-cells, T-cell clones, immature thymocytes and a proportion of B-cells, and is rapidly induced on naive T-cells after activation. -
