Mouse MMP9 ELISA Kit (ab253227) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of MMP9 protein in cell culture supernatant, hep plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse MMP9 with 38.91 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
MMP9 (gelatinase B) is a member of the matrix metalloproteinases family of zinc and calcium dependent endopeptidases. MMP9 is a 95kDa protein produced by keratinocytes, monocytes, macrophages and PMN leukocytes. It is able to degrade all the components of the extracellular matrix including gelatin, collagen types IV and V, elastin and proteoglycan core protein. MMP9 is believed to act synergistically with interstitial collagenase (MMP-1) in the degradation of fibrillar collagens as it degrades their denatured gelatin forms. MMP9 is present in most inflammatory responses. Mouse MMP9 shares 72.9% sequence homology with primate. 71.8% with human, 81.6 % with rat, and 73.0% with bovine MMP9.

The conserved cysteine present in the cysteine-switch motif binds the catalytic zinc ion, thus inhibiting the enzyme. The dissociation of the cysteine from the zinc ion upon the activation-peptide release activates the enzyme. -

The concentrations of MMP9 were measured in duplicates, interpolated from the MMP9 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 5%, plasma (citrate) 5% and plasma (heparin) 25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean MMP9 concentration was determined to be 158.72 ng/mL in serum and 37.15 ng/mL in plasma (heparin).

Undiluted samples are as follows: unstimulated P388D1 supernatant 100% and LPS stimulated P388 D1 10%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean MMP9 concentration was determined to be 1.03 ng/mL in unstimulated P388D1 supernatant and 74.58 ng/mL in LPS stimulated P388D1.
