Product overview
Mouse MAP2 ELISA Kit (ab253229) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of MAP2 protein in cell culture supernatant, cit plasma, edta plasma, hep plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse MAP2 with 43.31 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies
- Fully validated in biological samples
- 96-wells plate breakable into 12 x 8 wells stripsA 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

Notes
MAP2 (Microtubule-associated protein 2) is a neuronal protein that regulates the structure and stability of microtubules. It belongs to the MAP2/Tau family. Multiple MAP2 isoforms are expressed in neurons, including high molecular weight MAP2A and MAP2B (280 and 270 kDa), and low molecular weight MAP2C and MAP2D (70 and 75 kDa). Phosphorylation of MAP2 modulates its association with the cytoskeleton and is developmentally regulated. Mouse MAP2 shares 80.2%, 92.3%, and 74.4% sequence homology with human, rat, and bovine, respectively.

Function
The exact function of MAP2 is unknown but MAPs may stabilize the microtubules against depolymerization. They also seem to have a stiffening effect on microtubules. -
Sequence similarities
Contains 3 Tau/MAP repeats. -
Post-translational
modificationsPhosphorylated at serine residues in K-X-G-S motifs by MAP/microtubule affinity-regulating kinase (MARK1 or MARK2), causing detachment from microtubules, and their disassembly (By similarity). Isoform 2 is probably phosphorylated by PKA at Ser-323, Ser-354 and Ser-386 and by FYN at Tyr-67. -

SimpleStep ELISA technology allows the formation of the antibody-antigen complex in one single step, reducing assay time to 90 minutes. Add samples or standards and antibody mix to wells all at once, incubate, wash, and add your final substrate. See protocol for a detailed step-by-step guide.

The MAP2 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of MAP2 were measured in duplicate and interpolated from the MAP2 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean MAP2 concentration was determined to be 3632.84 pg/mL in mouse brain tissue extract.
