Mouse Endostatin ELISA Kit (ab263894) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Endostatin protein in cell culture supernatant, cit plasma, edta plasma, hep plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse Endostatin with 2.33 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
Endostatin is a cleaved product of the C-terminal domain of collagen XVIII, an endogenous angiogenesis inhibitor. Endostatin localized to wall of vasculature in the elastic fiber and basement membrane where it inhibits endothelial cell migration, in vivo and in vitro. Endostatin induces endothelial cell apoptosis and inhibits tumor growth and impairs blood vessel maturation in wound healing. It is implicated in endothelial cell adhesion and cytoskeletal organization. Mouse endostatin shares a 70, 67, 67% sequence homology with human, rat, and cow, respectively.

Relevance
Endostatin (20-22 kDa), a cleaved product of the C-terminal domain of collagen XVIII, is an endogenous angiogenesis inhibitor. It was originally found in conditioned media from a murine endothelial tumor cell line, hemangioendothelioma. Endostatin inhibits endothelial cell migration (in vivo and in vitro) and induces endothelial cell apoptosis. It inhibits tumor growth and impairs blood vessel maturation in wound healing. Endostatin has an important role in endothelial cell adhesion and cytoskeletal organization. Endostatin can be found in vessel walls (elastic fibers) and basement membranes. -

The concentrations of Endostatin were measured in duplicates, interpolated from the Endostatin standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 1:200, plasma (citrate) 1:200, plasma (EDTA) 1:200 and plasma (heparin) 1:200. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Endostatin concentration was determined to be 97.87 ng/mL in serum, 72.00 ng/mL in plasma (citrate), 85.87 ng/mL in plasma (EDTA), and 101.37 ng/mL in plasma (heparin).

Interpolated concentrations of native Endostatin in mouse cell and tissue culture supernatant samples.
The concentrations of Endostatin were measured in duplicates, interpolated from the Endostatin standard curves and corrected for sample dilution. Undiluted samples are as follows: heart supernatant 1:8 and kidney supernatant 1:200. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Endostatin concentration was determined to be 7.60 ng/mL in heart supernatant and 67.42 ng/mL in kidney supernatant.
