Mouse CYR61 ELISA Kit (ab253223) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of CYR61 protein in cell culture media, cell culture supernatant, edta plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse CYR61 with 10.1 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. CYR61 (Cysteine-rich Angiogenic Inducer 61), also known as CCN Family Member 1 (CCN1) or Insulin-Like Growth Factor-Binding Protein 10 (IGFBP10) is a secreted protein that promotes cell proliferation, chemotaxis, angiogenesis, and cell adhesion. CYR61 exhibits both tumorigenic and tumor suppressor properties, depending in part on the localization.

Function
Promotes cell proliferation, chemotaxis, angiogenesis and cell adhesion. Appears to play a role in wound healing by up-regulating, in skin fibroblasts, the expression of a number of genes involved in angiogenesis, inflammation and matrix remodeling including VEGA-A, VEGA-C, MMP1, MMP3, TIMP1, uPA, PAI-1 and integrins alpha-3 and alpha-5. CYR61-mediated gene regulation is dependent on heparin-binding. Down-regulates the expression of alpha-1 and alpha-2 subunits of collagen type-1. Promotes cell adhesion and adhesive signaling through integrin alpha-6/beta-1, cell migration through integrin alpha-v/beta-5 and cell proliferation through integrin alpha-v/beta-3. -

Interpolated concentrations of native CYR61 in mouse serum, plasma (EDTA), and cell culture supernatant samples.
The concentrations of CYR61 were measured in duplicates, interpolated from the CYR61 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 50%, plasma (EDTA) 100%, and NIH/3T3 supernatant 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean CYR61 concentration was determined to be 209.0 pg/mL in neat serum, 119.6 pg/mL in neat plasma (EDTA), and 137.3 pg/mL in neat NIH/3T3 cell culture supernatant.
