General notes
Limonene Mounting Medium ab104141 is an organic mounting medium compatible with chromogens such as DAB. Suitable for hematoxylin and eosin (H and E) staining.
This mounting medium is made with limonene, a natural product from orange peels. It is good for preserving tissues and cell smears that can be dehydrated with organic solvents in immunohistochemistry (IHC) e.g. DAB and DAB with nickel or cobalt. It is an excellent choice for mounting H and E stained slides.
Intended Use Mounting tissues, cell smears in Immunohistochemistry for chromogens that are resistant to organic solvents e.g. DAB, DAB with cobalt, DAB with Nickel any other chromogens that are resistant to organic solvents dehydration. H & E (hematoxylin and Eosin) staining.
Mounting media products
For fluorescent cell and tissue staining, Abcam recommends aqueous, anti-fade Fluorescence Mounting Medium ab104135, Mounting Medium with DAPI ab104139, and Mounting Medium with PI ab104129. For thick sections or tissues containing lots of fat, we recommend Glycerol Mounting Medium with DAPI ab188804.
For chromogenic immunohistochemistry with the organic solvent-soluble substrates AEC or Fast Red, we recommend aqueous Mounting Medium ab64230.
Procedure
- Dehydrate tissue or cell smear slides with dehydrating agents e.g. 30 to 100% alcohol followed by xylene, toluene or any other dehydrating reagents.
- Add 2-3 drops of mounting medium. Apply coverslip carefully, avoiding air bubbles.
- Visualize slide under a microscope. The slides can be dried by leaving at room temperature overnight or by heating at 37-40°C for several hours.
