Human TrkB ELISA Kit (ab264625) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of TrkB protein in tissue extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human TrkB with 23.76 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
TrkB, also known as NTRK2, is a tyrosine kinase receptor involved in the development of the peripheral and central nervous system. TrkB plays a role in synapse plasticity and formation, and neuron survival, proliferation and differentiation. Ligand (including BDNF and NTF4) interactions induces homodimerization, autophosphorylation and activation of TrkB. This kit targets an N-terminal fragment of the mature chain and shares approximately 90% sequence identity with mouse and rat.

Function
Receptor for brain-derived neurotrophic factor (BDNF), neurotrophin-3 and neurotrophin-4/5 but not nerve growth factor (NGF). Involved in the development and/or maintenance of the nervous system. This is a tyrosine-protein kinase receptor. Known substrates for the TRK receptors are SHC1, PI-3 kinase, and PLC-gamma-1. -
Tissue specificity
Isoform TrkB is widely expressed, mainly in the nervous tissue. In the CNS, expression is observed in the cerebral cortex, hippocampus, thalamus, choroid plexus, granular layer of the cerebellum, brain stem, and spinal cord. In the peripheral nervous system, it is expressed in many cranial ganglia, the ophtalmic nerve, the vestibular system, multiple facial structures, the submaxillary glands, and dorsal root ganglia. Isoform TrkB-T1 is expressed in multiple tissues, mainly in brain, pancreas, kidney and heart. Isoform TrkB-T-Shc is predominantly expressed in brain. -

The concentrations of TrkB were measured in duplicate and interpolated from the TrkB standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean TrkB concentration was determined to be 1,934 pg/mL in brain extract.
