Prekallikrein 1B in vitro CatchPoint SimpleStep ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of Prekallikrein and Plasma Kallikrein 1B protein in human serum, plasma, milk, and cell culture supernatant.

If using a Molecular Devices’ plate reader supported by SoftMax® Pro software, a preconfigured protocol for these CatchPoint SimpleStep ELISA Kits is available with all the protocol and analysis settings at www.softmaxpro.org.The CatchPoint SimpleStep ELISA employs an affinity tag labeled capture antibody and a reporter conjugated detector antibody which immunocapture the sample analyte in solution. This entire complex (capture antibody/analyte/detector antibody) is in turn immobilized via immunoaffinity of an anti-tag antibody coating the well. To perform the assay, samples or standards are added to the wells, followed by the antibody mix. After incubation, the wells are washed to remove unbound material. CatchPoint HRP Development Solution containing the Stoplight Red Substrate is added. During incubation, the substrate is catalyzed by HRP generating a fluorescent product. Signal is generated proportionally to the amount of bound analyte and the intensity is measured in a fluorescence plater reader at 530/570/590 nm Excitation/Cutoff/Emission.

Notes
Prekallikrein 1B (KLKB1) is a 88 kDa serine protease involved in blood coagulation, kinin generation, fibrinolysis and inflammation. Mature KLKB1 primarily circulates as a complex with high-molecular weight kinnogen, where it releases kinins from kininogens during the inflammation and vasodilation response.
This kit detects both prekallikrein and plasma kallikrein.

Function
The enzyme cleaves Lys-Arg and Arg-Ser bonds. It activates, in a reciprocal reaction, factor XII after its binding to a negatively charged surface. It also releases bradykinin from HMW kininogen and may also play a role in the renin-angiotensin system by converting prorenin into renin. -
Involvement in disease
Defects in KLKB1 are the cause of prekallikrein deficiency (PKK deficiency) [MIM:612423]; also known as Fletcher factor deficiency. This disorder is a blood coagulation defect. -

Serum from 10 healthy male donors was measured in duplicate. The mean Plasma Kallikrein 1B concentration was determined to be 112.6 µg/mL with a range of 80.8 – 206.4 µg/mL in male donors.
