Human MASP2 ELISA kit (ab278121) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human MASP2 protein in human serum and plasma. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human MASP2 with 21 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
MASP2, Mannan-binding lectin serine protease 2, plays a key role in the complement activation cascade. This protein is part of a larger mannose-binding complex which senses foreign material, activates C3 convertase, and perpetuates the immune response. The immunogen from which the antibodies in this kit were raised represents the N-terminal half of the protein. This protein does not share high sequence identity in mouse or rat samples (80 and 81%, respectively) and, therefore, this kit is not expected to cross-react.

MASP2, MBL associated serine protease 2, is a serum protease that plays an important role in the activation of the complement system via mannose-binding lectin (MBL). Binding of MBL to carbohydrate groups triggers MASP2 activation by auto-catalytic cleavage which leads to cleavage of C2 and C4, leading to their activation and to the formation of C3 convertase. MASP2 also binds to L-Ficolin and H-Ficolin, and when the complex binds pathogens the MASP is activated. Low MBL levels in serum are associated with increased susceptibility to infection, and an Asp105Gly mutation in MASP2 is associated with low MASP2 levels in serum. Reduced functional levels of MBL are also postulated to be associated with generation of autoantibodies in systemic lupus erythematosus, acting through a decreased clearance of apoptotic material. A truncated form of MASP2 that constitutes the amino 185 residues, Map19 is also a component of the MBL-MASP-1 complex. -

The MASP2 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of MASP2 were measured in duplicates, interpolated from the MASP2 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 1%, plasma (citrate) 1%, plasma (heparin) 1% and plasma (EDTA) 1%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean MASP2 concentration was determined to be 484.87 ng/mL in serum, 561.89 ng/mL in plasma (citrate), 500.49 ng/mL in plasma (heparin), and 404.43 ng/mL in plasma (EDTA).

Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean MASP2 concentration was determined to be 343.24 ng/mL with a range of 56.31 – 528.11 ng/mL.
