Human IL-17C ELISA kit (ab279410) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human IL-17C protein in human serum, plasma and cell culture supernatant samples. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human IL-17C with 1.2 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
IL-17C, also known as CX2, is a secreted cytokine of IL-17 family. IL-17C binds to a heterodimer formed by IL-17RA and IL-17RE. IL-17C plays a crucial role in innate immunity of the epithelium, including to intestinal bacterial pathogens, in an autocrine manner. It stimulates the production of antibacterial peptides and proinflammatory molecules for host defense by signaling through the NF-kappa-B and MAPK pathways. IL-17C is up-regulated by bacterial stimuli, including by lipopolysaccharides in gut epithelial cell lines, and by various proinflammatory cytokines, including TNF, IL-1B and IL-17F. IL-17C acts synergistically with IL-22 in inducing the expression of antibacterial peptides, and with TNF and IL-1B in inducing DEFB2 from keratinocytes. Depending on the type of insult, IL-17C may have both protective and pathogenic properties, either by maintaining epithelial homeostasis after an inflammatory challenge or by promoting inflammatory phenotype. It plays an important role in the pathogenesis of inflammatory arthritis.

The IL-17C standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of IL-17C were measured in duplicates, interpolated from the IL-17C standard curves and corrected for sample dilution. Undiluted samples are as follows: treated HCT 116 supernatant 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IL-17C concentration was determined to be 93.5 pg/mL in neat treated supernatant. There was no detectable signal in mock treated HCT 116 cell supernatant.
