Product overview
Human IL-13R alpha 2 ELISA kit (ab274400) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human IL-13R alpha 2 protein in serum, plasma cell culture supernatant, and cell and tissue extract. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human IL-13R alpha 2 with 96.3 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
-Single-wash protocol reduces assay time to 90 minutes or less
-High sensitivity, specificity and reproducibility from superior antibodies
-Fully validated in biological samples
-96-wells plate breakable into 12 x 8 wells strips
A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

Notes
Interleukin-13 receptor alpha 2 is one of the receptors for IL-13, which is a T helper 2 molecule involved in immune response, inflammation, and wound healing. Studies have shown overexpression of IL-13R alpha 2 can confer invasive and metastatic ability in variety of cancer cells such as melanoma, ovarian, pancreatic, colorectal, and gliomas. Therefore, making IL-13R alpha 2 a viable candidate as a prognostic biomarker. This kit is developed to recognize and target only IL-13R alpha 2.

Research areas
-
Function
Binds as a monomer with high affinity to interleukin-13 (IL13), but not to interleukin-4 (IL4). -
Sequence similarities
Belongs to the type I cytokine receptor family. Type 5 subfamily.
Contains 1 fibronectin type-III domain. -
Domain
The WSXWS motif appears to be necessary for proper protein folding and thereby efficient intracellular transport and cell-surface receptor binding.
The box 1 motif is required for JAK interaction and/or activation. -

SimpleStep ELISA technology allows the formation of the antibody-antigen complex in one single step, reducing assay time to 90 minutes. Add samples or standards and antibody mix to wells all at once, incubate, wash, and add your final substrate. See protocol for a detailed step-by-step guide.

The IL-13R alpha 2 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.
-
The concentrations of IL-13R alpha 2 were measured in duplicates, interpolated from the IL-13R alpha 2 standard curves and corrected for sample dilution. Undiluted samples are as follows: A375 supernatant neat. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IL-13R alpha 2 concentration was determined to be 3,741 pg/mL in A375 supernatant.

The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IL-13R alpha 2 concentration was determined to be 8,147 pg/mL in U-87MG cell extract and 10,842 pg/mL in A375 cell extract.
