Human HRG ELISA Kit (ab277404) is an in-vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human HRG in serum, plasma and cell culture supernatants.
This assay employs an antibody specific for Human HRG coated on a 96-well plate. Standards and samples are pipetted into the wells and Human HRG present in a sample is bound to the wells by the immobilized antibody. The wells are washed, and biotinylated anti-Human HRG antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Human HRG bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Relevance
HRG (histidine-rich glycoprotein) contains two cystatin-like domains and is located in plasma and platelets. The physiological function is not yet known. It binds heme, dyes and divalent metal ions. It can inhibit rosette formation and interacts with heparin, thrombospondin, and the lysine-binding site of plasminogen. A potential prothrombotic effect of HRG is indicated by the inhibition of fibrinolysis and the reduction of inhibition of coagulation. Mutations in HRG lead to thrombophilia due to abnormal histidine-rich glycoprotein levels. -
