Human Ghrelin ELISA Kit (Ghrelin-28) (ab263887) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Ghrelin (Ghrelin-28) protein in cell culture media, cit plasma, hep plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Ghrelin (Ghrelin-28) with 1.048 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
Ghrelin is a peptide hormone, it is a ligand of growth hormone secretagogue receptor type 1 (GHSR). It is involved in appetite stimulation and gastric acid secretion and induces adiposity. It is involved in growth regulation and induces the release of growth hormone from the pituitary. Ghrelin exists in two forms, Ghrelin-28 and one amino acid shorter Ghrelin-27. Both forms as well as Obestatin are generated by cleavage from common precursor protein, Appetite-Regulating Hormone coded by GHRL gene. Ghrelin-28 and Obestatin have non-overlapping amino acid sequences. O-octanoylation or O-decanoylation of Ghrelins is essential for Ghrelin activity.

Function
Ghrelin is the ligand for growth hormone secretagogue receptor type 1 (GHSR). Induces the release of growth hormone from the pituitary. Has an appetite-stimulating effect, induces adiposity and stimulates gastric acid secretion. Involved in growth regulation.
Obestatin may be the ligand for GPR39. May have an appetite-reducing effect resulting in decreased food intake. May reduce gastric emptying activity and jejunal motility. -
Tissue specificity
Highest level in stomach. All forms are found in serum as well. Other tissues compensate for the loss of ghrelin synthesis in the stomach following gastrectomy. -

Post-translational
modificationsO-octanoylation or O-decanoylation is essential for ghrelin activity. The O-decanoylated forms Ghrelin-27-C10 and Ghrelin-28-C10 differ in the length of the carbon backbone of the carboxylic acid bound to Ser-26. A small fraction of ghrelin, ghrelin-28-C10:1, may be modified with a singly unsaturated carboxylic acid.
Amidation of Leu-98 is essential for obestatin activity. -

The Ghrelin standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of Ghrelin were measured in duplicates, interpolated from the Ghrelin standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 50%, plasma (citrate) 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Ghrelin concentration was determined to be 25.69 pg/mL in neat serum and 31.66 pg/mL in neat plasma (citrate).
