Human FAP ELISA Kit (ab256404) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of FAP protein in cell culture media, cit plasma, edta plasma, hep plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human FAP with 14.96 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
FAP (fibroblast activation protein-alpha; also known as seprase) is a 86.3 kD type II transmembrane serine protease that degrades gelatin, Type I collagen, and N-terminal prolines. FAP is induced on reactive stromal fibroblasts in epithelial cancers, sarcomas, and granulation tissue. FAP may play roles in tumor invasion, tissue remodeling, and wound repair. Human FAP shares 90.1%, 89.5%, 93.9%, and 97.3% sequence homology with mouse, rat, bovine, and monkey, respectively.

The FAP standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of FAP were measured in duplicates, interpolated from the FAP standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 5%, plasma (citrate) 10%, plasma (EDTA) 10%, and plasma (heparin) 10%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean FAP concentration was determined to be 29.25 ng/mL in serum, 24.36 ng/mL in plasma (citrate), 26.29 ng/mL in plasma (EDTA), and 24.01 ng/mL in plasma (heparin).

Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean FAP concentration was determined to be 53.12 ng/mL with a range of 20.47 – 101.21 ng/mL.
