Human Desmoglein 3 ELISA kit (ab275902) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human Desmoglein 3 protein in human cell extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Desmoglein 3 with 8 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
Desmoglein 3 (DSG3, PVA) is a key component of cell-cell junctions in many epithelial type cells. Auto-antibody formation to DSG3 and subsequent loss of adhesive function is the main cause of pemphigus vulgaris, a skin blistering disease. Antibodies for this kit were raised against an immunogen which represents the N-terminal half of the mature protein. This protein does not share high sequence identity in mouse or rat samples (72 and 68%, respectively) and, therefore, this kit is not expected to cross-react.

The Desmoglein 3 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of Desmoglein 3 were measured in duplicate and interpolated from the Desmoglein 3 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Desmoglein 3 concentration was determined to be 2331.46 pg/mL in HACAT cell extract and 1827.74 pg/mL in A431 cell extract.
