Human CTHRC1 ELISA kit (ab274399) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human CTHRC1 protein in serum, plasma and cell culture supernatant. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human CTHRC1 with 27.3 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Notes
Collagen triple helix repeat containing protein 1 (CTHRC1) is a 30 kDa secreted glycoprotein encoded by the CTHRC1 gene. CTHRC1 is known to regulate extracellular collagen deposition and partake in vascular remodeling of arterial injury as a function of cellular response through the TGF-β and Wnt pathways. Moreover, mutation of the gene has been associated with carcinogenesis in a variety of cell and tissues, most notably esophageal adenocarcinoma and Barrett’s esophagus. Furthermore, recent studies have demonstrated CTHRC1 as a marker of the severity of the disease progression in the dystrophic muscles.

The CTHRC1 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of CTHRC1 were measured in duplicates, interpolated from the CTHRC1 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 50%, plasma (citrate) 50%, and SK-MEL28 cell culture supernatant 4%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean CTHRC1 concentration was determined to be 9,486 pg/mL in serum, 10,463 pg/mL in plasma (citrate) and 140,962 pg/mL in SK-MEL28 cell culture supernatant.
